At any given moment, each cell in the human body contains a certain amount of cell-free DNA (cfDNA), which in turn reflects its health status and disease risk for many serious diseases. Therefore, cfDNA has been proposed as a potential noninvasive biomarker for many diseases, including cancer, cardiovascular disease, diabetes, and neurodegenerative disorders. However, cfDNA is short-lived and degraded rapidly in the blood, limiting its clinical use. We propose the use of hydroxyapatite nanoparticles (HANs), which have good biocompatibility, to protect and stabilize circulating cfDNA, and as a potential carrier for targeted delivery of cfDNA to disease sites. In this study, we have developed a new HAN-based nanosized carrier for targeted delivery of cfDNA to disease sites, namely G3@SeHANs, which scavenges cfDNA and promotes osteogenic differentiation of cells through the AMPK signaling pathway. We also show that G3@SeHANs suppress alveolar bone loss in a mouse model of periodontitis, indicating that this cfDNA-scavenging approach could benefit the treatment of periodontitis.
